Study in vitro on hemoperfusion to eliminate chlorpyrifos and its metabolites

  • Objective To study the clearance of hemoperfusion in vitro against chlorpyrifos and its metabolites.
    Methods The experimental model for hemoperfusion in vitro was established. The effects of plasma concentrate, blood flow velocity, type of adsorbents and perfusion time on the clearance of chlorpyrifos and its metabolites were evaluated by orthogonal experimental design.
    Results The orthogonal experimental test showed that all the studied factors, including plasma concentration, blood flow velocity, type of adsorbents and perfusion time, significantly influenced the absorption of chlorpyrifos and its metabolites (P < 0.01). The order of factors affecting the absorption was perfusion time, type of adsorbents, blood toxins concentrate, and blood flow velocity. Orthogonal experiments showed that the best combination was 2 mL/min blood flow rate, activated carbon adsorbent and hemoperfusion time for 120 minutes for low blood concentration of chlorpyrifos (1 mg/L) and its metabolites (3, 5, 6-trichloro-2-pyridinol, 10 mg/L), and the clearance rate of chlorpyrifos and its metabolites was the highest.
    Conclusion In vitro test show that hemoperfusion could clear chlorpyrifos and its metabolites. Blood flow velocity, type of adsorbents and perfusion time could influence the absorption of chlorpyrifos.
